FAQ EC Blue Coliform and E Coli Detection in Water Samples

What type of water can we test with EC Blue?

EC blue can be used to test E. coli and coliform contamination in a broad range of water samples including drinking water, seawater, sewage water, chlorinated water (e.g. swimming pool) etc.

Is EC Blue compatible with chlorinated water?

Yes, EC Blue is suitable for chlorinated water analysis, the addition of sodium thiosulfate is recommended for water with a high concentration of chlorine.

How to discriminate coliform from E. coli using EC Blue?

The EC Blue formulation contains 2 different substrates: one chromogenic for the detection of coliforms and a fluorescent substrate that is specifically hydrolyse by E. coli.

Coliforms give a green/green-blue colouration after 24h of incubation at 37 °C and E. coli produce fluorescence, observable under UV-light.

Is EC Blue a qualitative or quantitative method?

The EC Blue product range can be used either qualitatively or quantitatively for the analysis of water.

EC Blue 100 (05593-EB0-080) and 100P (05591-EBP-100) provide a qualitative assessment of water contamination.

Quantitative measurement of bacterial contamination of water can be performed using EC Blue in combination with EC BlueQuant (06517-EBQ-018) using the Most Probable Number (MPN) method.

What is the shelf life of the EC Blue product range?

EC Blue 100 (05593-EB0-080) and 100P (05591-EBP-100) are stable for 24 months at room temperature (avoiding light exposure)

How to visually assess a positive sample?

Coliform positive sample displays a green to blue/green colour, visible to the eye.

E. coli positive samples are fluorescent after exposure to a UV light.

If interpretation is ambiguous, the EC Blue comparator (06514-EBC-001) will allow discrimination of positive and negative samples for both E. coli and coliform contamination.

How long do we need to incubate EC Blue?

Incubate samples at 37 °C for 24h.

What materials/equipment are required for food testing?

Food testing using CompactDry plates requires minimal lab equipment including pipettes, a sterile preparation area, and an incubator (a suitable homogeniser would be required for solid food homogenisation)

What is the difference between EC Blue 100P and EC Blue 100?

EC Blue 100P (05591-EBP-100) is a small pouch containing the required amount of culture media to allow the detection of E. coli and coliform in 100mL of tested water. The powder is added and solubilised directly into the test water sample followed by incubation.

EC Blue 100 (05593-EB0-080) is a sterile bottle containing the required amount of culture media to allow the detection of E.coli and coliforms. The test is performed by adding 100 mL of test water into the bottle and incubating.

How to dispose of EC Blue?

EC Blue bottles should be disposed according to local regulations for biohazardous materials.
